Serum from the 3rd immunization and pre-immunization was serially 2-fold diluted and added to ELISA plates coated with antigen proteins. Bound antibodies were detected using rabbit anti-VHH, followed by HRP conjugated goat anti-rabbit IgG (IRBS4012). TMB substrate was used for signal development, and the reaction was stopped with HCl. ELISA data were represented as the mean OD 450 nm ± SD from duplicate experiments.

The randomly selected expressed and purified VHH nanobodies were separated by SDS-PAGE under reducing conditions and visualized by PAS staining (lower panel). After immobilization on nitrocellulose, the membrane was blocked with 5% skim milk and probed with Rabbit Anti-Alpaca VHH (IRALP4502) at a 1:30,000 dilution, followed by detection with HRP-conjugated goat anti-rabbit (H+L) antibody (IRBS4012). Signals on the membrane were developed using ECL (right panel). Sample loading: 0.2 µg/well.
