The protein encoded by this gene is a protease inhibitor and cytokine transporter. It uses a bait-and-trap mechanism to inhibit a broad spectrum of proteases, including trypsin, thrombin and collagenase. It can also inhibit inflammatory cytokines, and it thus disrupts inflammatory cascades. Mutations in this gene are a cause of alpha-2-macroglobulin deficiency. This gene is implicated in Alzheimer's disease (AD) due to its ability to mediate the clearance and degradation of A-beta, the major component of beta-amyloid deposits. A related pseudogene, which is also located on the p arm of chromosome 12, has been identified. [provided by RefSeq, Nov 2016]

Anti- alpha 2-macroglobulin antibody [15A04-1G3] at 1/500 dilution
Lysates at 30 μg per lane.
This blot was produced using a 5% SDS-PAGE. The gel was run at 140V for 50 minutes before being transferred onto a Nitrocellulose membrane at 18V for 60 minutes. The membrane was then blocked for an hour before being incubated with IRM204 overnight at 4°C.

Immunofluorescence: cells were fixed with 4% paraformaldehyde for 10 min at RT, permeabilized with 0.1% NP-40 for 10 min at RT then blocked with 5% BSA for 30 min at room temperature. Cells were stained with IRM204 anti- alpha 2-macroglobulin antibody [15A04-1G3] (red) at 1:200 and 4°C. DAPI (blue) was used as the nuclear counter stain.
